This study developed and optimized a method for analyzing volatile organic compounds in urine headspace based on solid-phase microextraction, targeting portable gas chromatography-mass spectrometry systems. The research enhanced the method's sensitivity, repeatability, and chromatographic resolution by systematically optimizing off-column parameters such as sample volume, extraction temperature, and fiber coating type, as well as on-column and mass spectrometry parameters including desorption temperature, mass spectrometry scanning range, and column temperature program. The results showed that compared with the traditional probe sampling method, the solid-phase microextraction method enriched volatile organic compounds with an average log₂ fold change of 3.5, significantly increasing the number of detected volatile organic compounds. Moreover, the intra-day and inter-day relative standard deviations were mostly below 25%, indicating good repeatability. This portable system could distinguish the volatile organic compound profiles of urine from different healthy volunteers and standard urine, and its compound identification and quantitative results were verified by a benchtop gas chromatography-mass spectrometry system with high consistency. These findings demonstrate the potential application value of this method in clinical point-of-care detection of volatile organic compound biomarkers, providing new technical support for non-invasive disease diagnosis.
